The SHB protein also known as Src homology 2 domain-containing adapter protein B has a molecular mass of approximately 67 kDa. It functions as an adaptor protein mediating interactions between various signaling molecules. SHB facilitates signal transduction by forming connections between different proteins which can affect several cellular processes. This protein is expressed in a wide range of tissues including the liver pancreas and brain indicating its importance in multiple physiological contexts.
SHB plays a role in cellular responses to external stimuli and is involved in processes such as cell growth differentiation and apoptosis. It is a component of signal transduction pathways and works in conjunction with other signaling molecules to modulate these processes. SHB often forms complexes with other proteins allowing it to integrate signals from different pathways and coordinate complex cellular responses.
SHB participates in the MAPK/ERK and PI3K/AKT signaling pathways. These pathways are important for regulating cell survival growth and metabolism. SHB interacts with proteins like Grb2 and Sos within the MAPK/ERK pathway modulating their activity and in turn influencing downstream signaling events. In the PI3K/AKT pathway SHB associates with and modulates the activity of important mediator proteins further illustrating its integrative role in cellular signaling networks.
The dysregulation of SHB has been implicated in cancer and diabetes. In cancer SHB influences tumor progression and metastasis through its interactions with proteins like Src and PI3K which are critical in cancer biology. In diabetes altered SHB signaling affects insulin secretion and glucose metabolism linking it to diabetic pathophysiology. Understanding SHB's interactions in these diseases could provide insights into potential therapeutic targets.

Immunohistochemical analysis of paraffin-embedded mouse kidney and intestine tissues labeling SHB protein with ab308572 at 1/100. Antigen retrieval was performed using Citrate buffer, pH 6.0, 15 min
Various whole cell extracts (30 μg) were separated by 10% SDS-PAGE, and the membrane was blotted with SHB antibody HL1455 (ab308572) diluted at 1/1000. A HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody. Corresponding RNA expression data for the same cell lines are based on Human Protein Atlas program.
