SHCBP1 also known as SHC SH2-Domain Binding Protein 1 is a 72 kDa protein involved in cell cycle regulation. This protein exhibits a significant role in the interaction with the SHC1 adaptor protein influencing the signaling pathways activated by receptor tyrosine kinases. SHCBP1 shows high expression in tissues with active cell division such as the testis and several cancer cell lines. Its localization within cells includes both the cytoplasm and nucleus indicating diverse functional roles.
SHCBP1 aids in processes fundamental to cell proliferation and differentiation. It does not operate alone but participates in larger protein complexes that modulate cell division events. By interacting with other signaling molecules SHCBP1 becomes an important player in mitotic spindle assembly and cytokinesis. Its presence in complexes highlights its interaction with additional proteins which regulates the progression of the cell cycle.
SHCBP1 resides in pathways linked to the regulation of mitosis and cell proliferation. This protein plays an important role in the PI3K/Akt signaling pathway which is pivotal for cell growth and survival. Additionally ties to proteins like PLK1 and Aurora B further emphasize SHCBP1's integration into cell cycle regulation networks. These interactions ensure proper cellular responses to growth signals and accurate chromosome segregation during mitosis.
SHCBP1 connections to cancer and infertility are notable. In several types of cancer overexpression of SHCBP1 associates with tumor progression and poor prognosis often through its interaction with oncogenic proteins like Cyclin D1. In cases of male infertility dysregulation of SHCBP1 impacts spermatogenesis due to its essential role in cell cycle and division. SHCBP1 plays important roles in embryonic development, cytokinesis, neuronal development, tumorigenesis and T cell development. It is highly expressed in thymus but absent in liver. It can be induced in serum-stimulated NIH/3T3 cells. (PMID: 10086341 and 25153088).
