SHLD1 also known as shieldin complex subunit 1 functions as part of the shieldin complex involved in DNA repair. SHLD1 has a molecular mass of approximately 22 kDa. It is expressed in various tissues and is most abundant in cells that are actively dividing such as those found in cancer cell lines. SHLD1 plays a mechanical role by acting as a scaffold protein that contributes to the non-homologous end joining (NHEJ) pathway of DNA double-strand break repair.
Biological function summary

SHLD1 participates as a component of the shieldin complex alongside SHLD2 SHLD3 and REV7. Together these proteins facilitate the repair of DNA double-strand breaks by directing 53BP1-mediated NHEJ. This complex stabilizes the DNA ends and assists in their processing which prevents excessive resection and promotes the proper end-joining process.
