Supplementary information

SHQ1 is a protein involved in the processing and maturation of small nucleolar RNAs (snoRNAs). Also known as HSPC037 it has a molecular mass of approximately 65 kDa. SHQ1 is expressed in various tissues with its highest expression in the heart and skeletal muscle. It interacts directly with H/ACA ribonucleoproteins playing a role in the assembly and stability of these complexes which are essential for proper ribosome biogenesis.
Biological function summary

SHQ1 influences the proper assembly of H/ACA snoRNPs complexes important for the modification and maturation of ribosomal RNA (rRNA). SHQ1 primarily assists in the stabilization and assembly of dyskerin a catalytic subunit within H/ACA snoRNP complexes. Its function ensures accurate pseudouridylation a process that facilitates proper folding and functionality of rRNA within the ribosome. The protein's interaction within these complexes highlights its role in maintaining ribosomal integrity and function.

Pathways

The importance of SHQ1 lies within the snoRNA processing and ribosome assembly pathways. It plays a part in the ribosome biogenesis pathway which is vital for efficient protein synthesis. SHQ1 associates with dyskerin and other snoRNP proteins such as NOP10 and NHP2 linking them to the successful modification of rue rRNA. By influencing these interactions SHQ1 is integral to the proper synthesis and function of ribosomes.

Mutations or dysregulation of SHQ1 have been associated with disorders like X-linked Dyskeratosis Congenita and certain cancers. In Dyskeratosis Congenita defects in ribosome function arise due to inadequate rRNA pseudouridylation in which SHQ1 plays a significant part. Additionally anomalous expression of SHQ1 can lead to aberrant cell growth associating it with oncogenesis and tumor development. This connection to proteins such as dyskerin emphasizes the impact of SHQ1 in maintaining cellular health and its potential role in disease pathogenesis.

Target data

Required for the quantitative accumulation of H/ACA ribonucleoproteins (RNPs), including telomerase, probably through the stabilization of DKC1, from the time of its synthesis until its association with NOP10, NHP2, and NAF1 at the nascent H/ACA RNA.
