Catalyzes the removal of O-acetyl ester groups from position 9 of the free diacetylated sialate N-acetyl-9-O-acetylneuraminate (Neu5,9Ac2) in the cytosol and of the diacetylated sialate residues of sialylglycoconjugates in the lysosomes (Probable). Together with the sialate-O-acetyltransferase they regulate the balance of acetylated sialoglycoconjugates, key players in various processes such as cell-cell interactions, host-pathogen recognition, and tumor antigenicity (PubMed:21803834).

Sialic acid acetylesterase also known as SIAE is an enzyme that functions by removing acetyl groups from sialic acids on glycoconjugates. This enzyme belongs to the esterase family and exhibits a mass of approximately 60 kDa. SIAE shows expression in various tissues including the liver kidney and brain. It operates by breaking these acetyl linkages which is an important step in regulating sialic acid biology.
SIAE modifies the sialylation of glycoconjugates which impacts cellular recognition and communication. This enzyme is part of the immune regulation complex influencing how immune cells recognize and interact with each other as well as their environment. SIAE’s activity can alter cell surface properties which affects processes such as cell adhesion migration and signaling.
This enzyme appears in sialic acid metabolic pathways and immune response pathways. Within the context of the sialic acid metabolism pathway it works collaboratively with other enzymes like NEU1 and ST6GAL1 impacting the breakdown and synthesis of sialic acid derivatives. Additionally within immune response pathways SIAE influences the function of proteins involved in the regulation of immune cell signaling thereby affecting adaptive and innate immunity.
Researchers have connected SIAE with autoimmune diseases and certain neurodevelopmental disorders. Mutations or dysregulation in SIAE have been linked to conditions like systemic lupus erythematosus (SLE) and autism spectrum disorder (ASD). In the context of SLE SIAE's interaction with proteins like CTLA-4 impacts immune tolerance and autoreactivity. In ASD abnormal sialic acid modifications have implications for neurodevelopmental processes.
