Supplementary information

SIGLEC15 also known as sialic acid-binding Ig-like lectin 15 is a transmembrane protein with an approximate mass of 50 kDa. This protein is a member of the SIGLEC family that binds sialic acids on glycoproteins and glycolipids. SIGLEC15 is highly expressed in cells of the osteoclast lineage and some other immune cells. It typically functions as a receptor that can modulate immune cell interactions and signaling by recognizing and binding to sialylated glycans on other cells.
Biological function summary

SIGLEC15 plays roles in modulating the immune system and bone remodeling processes. It is not usually part of a larger complex but primarily acts as a cell surface receptor. SIGLEC15 helps regulate osteoclast differentiation and activity which are important for maintaining bone homeostasis. It modulates immune responses by interacting with other immune cells which can dampen or activate immune signaling pathways based on the cellular context.

Pathways

SIGLEC15 influences both immune signaling and bone metabolism pathways. It is involved in the RANKL-mediated osteoclastogenesis pathway which is critical for the formation and activation of osteoclasts. Within these pathways SIGLEC15 can relate to other proteins like RANK and RANKL that are essential in osteoclast development and function. Through its expression and interactions SIGLEC15 plays key roles in the regulation of immune and skeletal systems.

SIGLEC15 plays a significant role in osteoporosis and certain cancers. In osteoporosis SIGLEC15 expression in osteoclasts is elevated which can lead to increased bone resorption and weakened bones. In cancer its expression on tumor-associated macrophages and other cells can help tumors evade immune surveillance. In such conditions proteins like RANK RANKL and other immune checkpoint proteins may also be implicated providing potential targets for therapeutic intervention alongside SIGLEC15.
