SIK1 SIK2 and SIK3 also known as salt-inducible kinases are serine/threonine protein kinases that play a role in cellular signaling. These proteins have molecular weights of approximately 75 kDa 105 kDa and 145 kDa respectively. SIK1 SIK2 and SIK3 share structural similarities but differ in their tissue expression. SIK1 is mostly found in the heart and liver SIK2 in adipose tissue and SIK3 shows widespread expression including the brain and skeletal muscle. Mechanically these kinases modify other proteins through phosphorylation influencing cellular activity.
Biological function summary

These kinases regulate various processes essential for maintaining cellular homeostasis. They are not part of any well-known complex but interact with several cellular components. SIKs influence gene expression by inhibiting transcriptional regulators and also regulate metabolism and cell survival. Their action impacts how cells respond to external stimuli largely through modification of transcriptional activity in response to hormonal signals and nutrient availability.

Pathways

SIK1 SIK2 and SIK3 are integral to the AMP-activated protein kinase (AMPK) signaling pathway and the cAMP response element-binding protein (CREB) pathway. These pathways control energy balance and respond to stress conditions. SIKs act alongside AMPK to modulate cellular energy status and also influence the CREB signaling pathway by impacting phosphorylation state. Regulatory relationships exist with proteins like 14-3-3 and HDACs which themselves are key players in cellular metabolism and gene expression.

These kinases have links to metabolic diseases and cancer. SIKs have been associated with obesity due to their role in adipocyte differentiation and lipid metabolism. They also relate to pancreatic cancer where altered kinase activity disrupts normal cell function and proliferation. In cancer SIKs interact with proteins involved in proliferation pathways including mTOR affecting tumor growth and progression.

Serine/threonine-protein kinase that plays a role in many biological processes such as fatty acid oxidation, autophagy, immune response or glucose metabolism (PubMed : 23322770, PubMed : 26983400). Phosphorylates 'Ser-794' of IRS1 in insulin-stimulated adipocytes, potentially modulating the efficiency of insulin signal transduction. Inhibits CREB activity by phosphorylating and repressing TORCs, the CREB-specific coactivators (PubMed : 15454081). Phosphorylates EP300 and thus inhibits its histone acetyltransferase activity (PubMed : 21084751, PubMed : 26983400). In turn, regulates the DNA-binding ability of several transcription factors such as PPARA or MLXIPL (PubMed : 21084751, PubMed : 26983400).
