Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-SIK2 antibody (AB53423)

IHC image of ab53423 staining in human hippocampus formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab53423, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

SIK2 also known as Salt-Inducible Kinase 2 is a protein kinase involved in various cellular processes. It has a molecular mass of approximately 94 kDa. SIK2 is expressed in many tissues with high levels found in adipose tissue liver and skeletal muscle. As a kinase it phosphorylates specific target proteins regulating their activity and influencing cellular mechanisms.
Biological function summary

SIK2 participates in energy metabolism and cellular response to external stress. It interacts with several co-factors to form active complexes facilitating its activity. This protein regulates lipid and carbohydrate metabolism therefore playing a significant role in maintaining cellular energy homeostasis. Additionally SIK2 has a function in controlling the inflammatory response providing a link between metabolism and immune function.

Pathways

SIK2 acts as an essential mediator in the AMPK signaling pathway. This pathway is important for energy balance within cells and is related to other kinases like AMPK and other SIK family members. SIK2 also contributes to the regulation of the cAMP signaling pathway which is linked to various physiological processes. Through these pathways SIK2 influences processes such as glucose uptake and fat oxidation and interacts with proteins like CREB and HDACs.

SIK2 has implications in conditions like obesity and type 2 diabetes. Dysregulation of SIK2 activity can lead to metabolic imbalance and insulin resistance. This kinase is also associated with cancer progression particularly in ovarian cancer where it influences cell proliferation. In these diseases SIK2's connection to proteins such as AMPK and CREB suggests a complex role in disease modulation and potential therapeutic targeting.

Serine/threonine-protein kinase that plays a role in many biological processes such as fatty acid oxidation, autophagy, immune response or glucose metabolism (PubMed : 23322770, PubMed : 26983400). Phosphorylates 'Ser-794' of IRS1 in insulin-stimulated adipocytes, potentially modulating the efficiency of insulin signal transduction. Inhibits CREB activity by phosphorylating and repressing TORCs, the CREB-specific coactivators (PubMed : 15454081). Phosphorylates EP300 and thus inhibits its histone acetyltransferase activity (PubMed : 21084751, PubMed : 26983400).
