The SIR2 protein also known as Silent mating type Information Regulation 2 homolog and sometimes called SIRT1 in humans serves as an NAD-dependent deacetylase. It weighs about 120 kDa and is expressed in various tissues including the brain heart and liver. Mechanically SIR2 removes acetyl groups from lysine residues on histones and other proteins influencing chromatin structure and gene expression. Its deacetylase activity plays a role in numerous cellular processes like DNA repair transcription and cell survival.
Biological function summary

SIR2 influences processes related to aging and metabolic regulation. It functions as a core component of the sirtuin protein family and associates with a complex that involves other sirtuins like SIRT2 and SIRT3. This protein alters the acetylation status of histones and non-histone proteins impacting cellular aging and stress resistance. By regulating mitochondrial biogenesis and oxidative stress responses SIR2 has critical roles in metabolic homeostasis and lifespan extension in model organisms.
