Biological function summary
SIR2 influences processes related to aging and metabolic regulation. It functions as a core component of the sirtuin protein family and associates with a complex that involves other sirtuins like SIRT2 and SIRT3. This protein alters the acetylation status of histones and non-histone proteins impacting cellular aging and stress resistance. By regulating mitochondrial biogenesis and oxidative stress responses SIR2 has critical roles in metabolic homeostasis and lifespan extension in model organisms.
Pathways
SIR2 participates actively in metabolic and stress-response pathways. It plays a significant role in the NAD+ salvage pathway and is implicated in the insulin/IGF-1 signaling pathway. SIR2 modulates the activity of several key proteins including the transcription factors FOXO and p53. FOXO proteins regulated by SIR2 deacetylation mediate the oxidative stress response and affect glucose metabolism. Interaction with p53 results in modulation of cell cycle arrest and apoptosis demonstrating SIR2’s influence beyond metabolism.
