SIX2 also known as Sine Oculis Homeobox Homolog 2 is a transcription factor with a molecular weight of approximately 33 kDa. It contains a homeobox domain that allows binding to specific DNA sequences to regulate gene expression. SIX2 is mainly expressed in the developing kidney and plays an essential role during organogenesis. It is also found in some adult tissues but at lower levels compared to development stages.
Biological function summary

SIX2 influences cell proliferation and differentiation mostly during early kidney development. It partners with co-activators and is an integral component of developmental signaling complexes. In this context SIX2 helps maintain progenitor cell populations in an undifferentiated state which is vital for proper organ formation and function.

Pathways

SIX2 plays an essential role in pathways like Wnt and Pax signaling. These pathways regulate important developmental processes by interacting with proteins such as TCF/LEF and PAX2. Through these interactions SIX2 contributes to controlling cell fate decisions and organ patterning during embryogenesis ensuring proper organ development.

SIX2 is associated with renal agenesis and certain types of congenital abnormalities affecting the kidney and urinary tract. Disruptions in the SIX2 expression or function might lead to the absence of kidney formation. It is also linked to pathological conditions interacting with proteins such as GDNF and RET which can contribute to irregular kidney development or diseases involving kidney hyperplasia.

Target data

Transcription factor that plays an important role in the development of several organs, including kidney, skull and stomach. During kidney development, maintains cap mesenchyme multipotent nephron progenitor cells in an undifferentiated state by opposing the inductive signals emanating from the ureteric bud and cooperates with WNT9B to promote renewing progenitor cells proliferation. Acts through its interaction with TCF7L2 and OSR1 in a canonical Wnt signaling independent manner preventing transcription of differentiation genes in cap mesenchyme such as WNT4. Also acts independently of OSR1 to activate expression of many cap mesenchyme genes, including itself, GDNF and OSR1. During craniofacial development plays a role in growth and elongation of the cranial base through regulation of chondrocyte differentiation. During stomach organogenesis, controls pyloric sphincter formation and mucosal growth through regulation of a gene network including NKX2-5, BMPR1B, BMP4, SOX9 and GREM1. During branchial arch development, acts to mediate HOXA2 control over the insulin-like growth factor pathway. May also be involved in limb tendon and ligament development (By similarity). Plays a role in cell proliferation and migration.
