SKA2 also known as spindle and kinetochore-associated protein 2 is an important component in the regulation of cell division. It has a mass of approximately 18 kDa and is expressed in tissues with high proliferative capacity such as the brain and liver. Mechanically SKA2 interacts with microtubules at the mitotic spindle and aids in the stabilization of kinetochore-microtubule attachments during mitosis. This interaction ensures proper chromosome segregation which is essential for successful cell division.
Biological function summary

This protein is part of the SKA complex which also includes SKA1 and SKA3. This complex stabilizes spindle fibers to enhance the attachment of chromosomes during mitosis. The SKA complex functions to maintain chromosomal alignment and ensure that sister chromatids separate correctly. Disruption of SKA2's role in this process can lead to chromosomal instability a factor often associated with various pathological conditions.

Pathways

SKA2 plays a significant role in the cell cycle and spindle assembly checkpoint pathways. The spindle assembly checkpoint ensures that cells do not proceed to anaphase until all chromosomes are properly aligned. SKA2 is closely related to the Ndc80 complex in these pathways. Proper function of these pathways is critical for maintaining genomic integrity and preventing aneuploidy a condition where cells have an abnormal number of chromosomes.

SKA2 has gathered attention due to its association with cancer and psychiatric disorders. Dysregulation of SKA2 contributes to tumorigenesis as it can lead to uncontrolled cell proliferation and chromosomal missegregation. In psychiatric contexts altered SKA2 expression has been linked to stress-related disorders including depression. SKA2 interacts with proteins such as FKBP5 in these disorders influencing glucocorticoid receptor sensitivity and stress response.

Target data

Component of the SKA1 complex, a microtubule-binding subcomplex of the outer kinetochore that is essential for proper chromosome segregation (PubMed : 17093495, PubMed : 19289083, PubMed : 23085020). Required for timely anaphase onset during mitosis, when chromosomes undergo bipolar attachment on spindle microtubules leading to silencing of the spindle checkpoint (PubMed : 17093495). The SKA1 complex is a direct component of the kinetochore-microtubule interface and directly associates with microtubules as oligomeric assemblies (PubMed : 19289083). The complex facilitates the processive movement of microspheres along a microtubule in a depolymerization-coupled manner (PubMed : 17093495, PubMed : 19289083). In the complex, it is required for SKA1 localization (PubMed : 19289083). Affinity for microtubules is synergistically enhanced in the presence of the ndc-80 complex and may allow the ndc-80 complex to track depolymerizing microtubules (PubMed : 23085020).
