Required for optimal conjugation between T-cells and antigen-presenting cells by promoting the clustering of integrin ITGAL on the surface of T-cells. Activity summary

SKAP55 also known as Src kinase-associated phosphoprotein 55 is a protein with a mass of approximately 55 kDa. It plays an important role in T-cell signaling. This protein mainly expresses in lymphoid tissues including the thymus spleen and peripheral blood lymphocytes. SKAP55 interacts with signaling molecules via its SH3 domain facilitating cellular communication and activation.

Biological function summary

The protein functions as an adapter molecule in T-cell receptor (TCR) signaling pathways. SKAP55 associates with the LAT-Gads complex contributing to intracellular signal transduction. It helps mediate actin cytoskeletal rearrangements necessary for immune synapse formation. These rearrangements are critical for effective immune response meaning SKAP55 is integral to T-cell activation and function.

Pathways

SKAP55 participates in the signaling events downstream of the TCR particularly in the PI3K and Ras-MEK-ERK pathways. It works closely with proteins like LCK and ZAP70 which are essential for the phosphorylation processes involved in T-cell activation. Through these interactions SKAP55 influences the organization of signaling assemblies promoting effective cellular responses.

Associated diseases and disorders

SKAP55 relates to immune system-related conditions like autoimmune diseases and immunodeficiencies. Alterations in SKAP55 expression or function can affect T-cell signaling potentially leading to improper immune responses. SKAP55 interactions with LCK can influence diseases such as systemic lupus erythematosus where immune regulation is disrupted highlighting its importance in maintaining immune system balance.
