Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-SLC12A6/KCC3 antibody - C-terminal (AB191451)
Immunohistochemical analysis of paraffin-embedded Human colorectal adenocarcinoma tissue labeling SLC12A6/KCC3 with ab191451 at 2 μg/ml. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with ab191451 overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. DAB was used as the chromogen.

Western blot - Anti-SLC12A6/KCC3 antibody - C-terminal (AB191451)
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with ab191451 at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1/5000 for 1.5 hour at RT.

KCC3 impacts cellular processes by being an important part of regulating salt balance within cells. It belongs to the cation-chloride cotransporter family and plays an important role in cell volume regulation. KCC3 does not usually function as part of a larger protein complex but acts as a standalone transporter. It helps stabilize cell environment which is necessary for correct cellular signaling and function.
Pathways
KCC3 is integral to maintaining the proper function of the WNK-SPAK/OSR1 kinase pathway. This pathway regulates ion transport and adjusts cell volume after osmotic stress. KCC3 interacts with proteins from this pathway like WNK kinases and SPAK which phosphorylate KCC3 to control its activity. As the transporter modifies ion flow it indirectly affects other signaling pathways dependent on ionic balance within cells.
