Mitochondrial sodium/calcium antiporter that mediates sodium-dependent calcium efflux from mitochondrion, by mediating the exchange of 3 sodium ions per 1 calcium ion (PubMed:15060069, PubMed:20018762, PubMed:22829870, PubMed:23056385, PubMed:24898248, PubMed:28130126, PubMed:28219928). Plays a central role in mitochondrial calcium homeostasis by mediating mitochondrial calcium extrusion: calcium efflux is essential for mitochondrial function and cell survival, notably in cardiomyocytes (By similarity). Regulates rates of glucose-dependent insulin secretion in pancreatic beta-cells during the first phase of insulin secretion: acts by mediating efflux of calcium from mitochondrion, thereby affecting cytoplasmic calcium responses (PubMed:23056385). Required for store-operated Ca(2+) entry (SOCE) and Ca(2+) release-activated Ca(2+) (CRAC) channel regulation: sodium transport by SLC8B1 leads to promote calcium-shuttling that modulates mitochondrial redox status, thereby regulating SOCE activity (PubMed:28219928). Involved in B-lymphocyte chemotaxis (By similarity). Able to transport Ca(2+) in exchange of either Li(+) or Na(+), explaining how Li(+) catalyzes Ca(2+) exchange (PubMed:15060069, PubMed:28130126). In contrast to other members of the family its function is independent of K(+) (PubMed:15060069).

SLC24A6 also known as the sodium/potassium/calcium exchanger 6 or NCKX6 is a membrane protein involved in the exchange of ions. This protein enables the transport of Na+ Ca2+ and K+ ions across cellular membranes. Its molecular mass is approximately 73 kDa. SLC24A6 is expressed in several tissues particularly in the brain where its function is important for maintaining ion balance within neuronal cells.
There are important roles that SLC24A6 plays in cellular homeostasis and signal transduction. As part of a complex network of ion transporters SLC24A6 aids in regulating calcium ion concentration within cells. This regulation is essential for neuronal signaling and overall cellular communication. The ability to exchange ions efficiently allows cells to respond accurately to physiological changes.
SLC24A6 is involved in important signaling cascades such as the calcium signaling pathway and calcineurin signaling pathway. In the calcium signaling pathway it helps modulate intracellular calcium levels which influence various cellular processes like muscle contraction and neurotransmitter release. SLC24A6 also interacts with other ion exchangers and transporters playing a role alongside proteins such as calmodulin which further affect neuronal function.
SLC24A6 has been associated with conditions like bipolar disorder and schizophrenia. Studies suggest that dysfunction in ion homeostasis due to altered SLC24A6 activity can lead to these neurological disorders.
