Catalyzes the transport of 2-oxoglutarate (alpha-oxoglutarate) across the inner mitochondrial membrane in an electroneutral exchange for malate. Can also exchange 2-oxoglutarate for other dicarboxylic acids such as malonate, succinate, maleate and oxaloacetate, although with lower affinity. Contributes to several metabolic processes, including the malate-aspartate shuttle, the oxoglutarate/isocitrate shuttle, in gluconeogenesis from lactate, and in nitrogen metabolism (By similarity). Maintains mitochondrial fusion and fission events, and the organization and morphology of cristae (PubMed:21448454). Involved in the regulation of apoptosis (By similarity). Helps protect from cytotoxic-induced apoptosis by modulating glutathione levels in mitochondria (By similarity).

The SLC25A11 protein also known as the mitochondrial 2-oxoglutarate/malate carrier protein plays an important role in transporting substrates across the inner mitochondrial membrane. It specifically facilitates the exchange of 2-oxoglutarate and malate between the mitochondria and the cytosol. SLC25A11 has a molecular mass of approximately 33 kDa. Expression of SLC25A11 occurs across various tissues with significant levels in the liver and skeletal muscle where high energy turnover is required.
SLC25A11 is integral to mitochondrial function and energy metabolism. It is part of a transporter complex involved in the tricarboxylic acid (TCA) cycle supporting the conversion of nutrients into energy. This process is essential for maintaining the cellular energy levels needed for various physiological functions. The efficient transport of 2-oxoglutarate and malate helps in balancing the redox and carbon status in cells contributing to overall cellular homeostasis.
SLC25A11 is essential in the TCA cycle and the malate-aspartate shuttle. These pathways are vital for energy production and the regulation of cellular metabolism. In the TCA cycle SLC25A11 works closely with other proteins like isocitrate dehydrogenase participating in the decarboxylation and oxidation processes. In the malate-aspartate shuttle SLC25A11 collaborates with malate dehydrogenase enabling effective NADH transfer into the mitochondria a critical step for ATP generation.
SLC25A11 is linked to metabolic and mitochondrial disorders. Mutations or dysfunction in SLC25A11 can contribute to conditions such as mitochondrial diseases where energy production is impaired. This protein also shows associations with some forms of cancer where altered metabolism is a hallmark. Connections with other proteins like fumarate hydratase highlight its role in pathways that when disrupted may lead to disease progression.
