Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of Human breast carcinoma tissue sections labeling SLC39A6/ZIP-6 with ab227696 at 1/100 dilution (4.05 µg/ml). Heat mediated antigen retrieval was performed Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution 2) for 10 mins. Rabbit specific IHC polymer detection kit HRP/DAB (ab209101) was used as the secondary antibody. Negative control : PBS instead of the primary antibody. Hematoxylin was used as a counterstain.

SLC39A6 also known as ZIP-6 functions as a zinc transporter involved in importing zinc into cells from the extracellular environment. This protein is part of the ZIP family and has a mass of approximately 93 kDa. ZIP-6 is frequently expressed in tissues such as liver kidney and various epithelial cells. ZIP transporters like ZIP-6 play an important role in zinc homeostasis by facilitating cellular uptake of this essential trace element which impacts many cellular processes.
Biological function summary

ZIP-6 actively regulates intracellular zinc levels essential for many cellular functions like growth and development. It exists independently and does not directly form a part of a larger complex. ZIP-6 enhances zinc-dependent signaling pathways and is necessary for different cellular tasks like gene expression and enzyme activity. ZIP-6's regulation of zinc concentrations affects the MAPK pathway which includes proteins like ERK1/2 highlighting its involvement in signal transduction and cellular response to stimuli.

ZIP-6 plays an essential role in cancer progression and immune system dysfunctions. Overexpression of ZIP-6 associates with breast cancer where it can promote proliferation and metastasis by affecting zinc-dependent pathways. ZIP-6 activity relates also to immune-related disorders influencing the inflammatory response through interactions with NF-kappaB. Changes in the activity or expression of ZIP-6 alter zinc homeostasis adding complexity to its disease-related impact.

Zinc-influx transporter which plays a role in zinc homeostasis and in the induction of epithelial-to-mesenchymal transition (EMT) (PubMed : 12839489, PubMed : 18272141, PubMed : 21422171, PubMed : 23919497, PubMed : 27274087, PubMed : 34394081). When associated with SLC39A10, the heterodimer formed by SLC39A10 and SLC39A6 mediates cellular zinc uptake to trigger cells to undergo epithelial- to-mesenchymal transition (EMT) (PubMed : 27274087). The SLC39A10-SLC39A6 heterodimer also controls NCAM1 phosphorylation and its integration into focal adhesion complexes during EMT (By similarity). Zinc influx inactivates GSK3B, enabling unphosphorylated SNAI1 in the nucleus to down-regulate adherence genes such as CDH1, causing loss of cell adherence (PubMed : 23919497). In addition, the SLC39A10-SLC39A6 heterodimer plays an essentiel role in initiating mitosis by importing zinc into cells to initiate a pathway resulting in the onset of mitosis (PubMed : 32797246).
