Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of Human breast carcinoma tissue sections labeling SLC39A6/ZIP-6 with ab227696 at 1/100 dilution (4.05 µg/ml). Heat mediated antigen retrieval was performed Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution 2) for 10 mins. Rabbit specific IHC polymer detection kit HRP/DAB (ab209101) was used as the secondary antibody. Negative control : PBS instead of the primary antibody. Hematoxylin was used as a counterstain.

ZIP-6 actively regulates intracellular zinc levels essential for many cellular functions like growth and development. It exists independently and does not directly form a part of a larger complex. ZIP-6 enhances zinc-dependent signaling pathways and is necessary for different cellular tasks like gene expression and enzyme activity. By controlling zinc entry into cells ZIP-6 supports several zinc-driven biological processes impacting cellular metabolism and survival.
Pathways
Researchers link ZIP-6 to the JAK-STAT signaling and MAPK pathways. In these pathways zinc acts as a necessary cofactor for numerous enzymes and transcription factors. ZIP-6's zinc transport activity connects with proteins such as STAT3 in the JAK-STAT pathway influencing cell proliferation and differentiation. ZIP-6's regulation of zinc concentrations affects the MAPK pathway which includes proteins like ERK1/2 highlighting its involvement in signal transduction and cellular response to stimuli.
