SLC40A1 also known as ferroportin or solute carrier family 40 member 1 is a transmembrane protein which functions as an iron transporter. This protein plays a role in exporting iron from cells into the bloodstream. The molecular weight of SLC40A1 is approximately 62 kDa. It is highly expressed in cells involved in iron storage and metabolism such as enterocytes in the duodenum and macrophages which process senescent red blood cells.
Biological function summary

The function of this transporter is important for iron homeostasis in human body. SLC40A1 helps balance iron levels by moving iron across intestinal cells into the plasma. It does not function as part of a larger protein complex but its activity is controlled by hepcidin a hormone that degrades SLC40A1 and regulates iron flow into the circulation.

Pathways

SLC40A1 is essential in the iron homeostasis pathway and the hepcidin regulation pathway. These pathways ensure iron supply matches body demands. It is closely related to hepcidin due to their interactive roles in this regulatory system. Hepcidin binds to SLC40A1 leading to its internalization and degradation which decreases iron export.

Mutations in SLC40A1 can lead to diseases like hemochromatosis characterized by excessive iron accumulation and anemia as iron transport becomes inefficient. Through these conditions SLC40A1 connects to hepcidin where disruption in their interaction contributes to disease pathology. Managing SLC40A1 activity is therefore important for maintaining proper iron levels and preventing related health issues.

Transports Fe(2+) from the inside of a cell to the outside of the cell, playing a key role for maintaining systemic iron homeostasis (PubMed : 15692071, PubMed : 22178646, PubMed : 22682227, PubMed : 24304836, PubMed : 29237594, PubMed : 29599243, PubMed : 30247984). Transports iron from intestinal, splenic, hepatic cells, macrophages and erythrocytes into the blood to provide iron to other tissues (By similarity). Controls therefore dietary iron uptake, iron recycling by macrophages and erythrocytes, and release of iron stores in hepatocytes (By similarity). When iron is in excess in serum, circulating HAMP/hepcidin levels increase resulting in a degradation of SLC40A1, thus limiting the iron efflux to plasma (PubMed : 22682227, PubMed : 29237594, PubMed : 32814342).
