Sodium/bicarbonate cotransporter which plays an important role in regulating intracellular pH (PubMed:10993873, PubMed:18319254). Has been shown to act as a sodium/bicarbonate cotransporter in exchange for intracellular chloride (By similarity). Has also been shown to act as a sodium/biocarbonate cotransporter which does not couple net influx of bicarbonate to net efflux of chloride, with the observed chloride efflux being due to chloride self-exchange (PubMed:18319254). Controls neuronal pH and may contribute to the secretion of cerebrospinal fluid (By similarity). Reduces the excitability of CA1 pyramidal neurons and modulates short-term synaptic plasticity (By similarity). Required in retinal cells to maintain normal pH which is necessary for normal vision (By similarity). In the kidney, likely to mediate bicarbonate reclamation in the apical membrane of the proximal tubules (By similarity).

SLC4A10 also known as NCBE (Na+-driven Cl-/HCO3- exchanger) is a member of the SLC4 family of bicarbonate transporters. It has a molecular mass of approximately 121 kDa. This protein functions in the exchange of bicarbonate (HCO3-) and chloride (Cl-) ions in cells and tissues facilitating the regulation of intracellular pH and bicarbonate homeostasis. SLC4A10 is strongly expressed in the brain particularly in structures like the cerebellum and hippocampus and is also present in other tissues such as the eyes and intestines. Its expression pattern indicates its role in specialized physiological processes requiring ion balance.
The functions of SLC4A10 are integral to neuronal ion balance and signaling. It transports ions across membranes contributing to the maintenance of pH in neural tissues important for neurotransmission and neuronal excitability. SLC4A10 does not function as part of a larger protein complex but interacts with other ion transport systems to maintain cellular ion homeostasis. Its activity ensures neurons efficiently communicate and respond to stimuli by modulating synaptic activity and plasticity supporting normal brain functions.
SLC4A10 plays roles in acid-base balance and ion transport pathways. It contributes significantly to the bicarbonate reclamation pathway which maintains pH balance within the central nervous system. This exchanger closely interacts with other transport proteins like SLC4A3/Band 3 and SLC4A7 to coordinate bicarbonate transport. SLC4A10 has associations with neurological conditions such as epilepsy and migraine. Studies have highlighted that dysfunctional SLC4A10 may lead to improper ion homeostasis which can predispose individuals to these disorders. Mutations or dysregulation of this protein can disrupt the ion balance within neurons leading to increased susceptibility to seizures and migraines. In the context of these disorders SLC4A10 may interact with other neuronal proteins such as the GABA transporter SLC6A1 affecting inhibitory signaling pathways and contributing to pathological neuronal excitability.
