SLC4A10 also known as NCBE (Na+-driven Cl-/HCO3- exchanger) is a member of the SLC4 family of bicarbonate transporters. It has a molecular mass of approximately 121 kDa. This protein functions in the exchange of bicarbonate (HCO3-) and chloride (Cl-) ions in cells and tissues facilitating the regulation of intracellular pH and bicarbonate homeostasis. SLC4A10 is strongly expressed in the brain particularly in structures like the cerebellum and hippocampus and is also present in other tissues such as the eyes and intestines. Its expression pattern indicates its role in specialized physiological processes requiring ion balance.
Biological function summary

The functions of SLC4A10 are integral to neuronal ion balance and signaling. It transports ions across membranes contributing to the maintenance of pH in neural tissues important for neurotransmission and neuronal excitability. SLC4A10 does not function as part of a larger protein complex but interacts with other ion transport systems to maintain cellular ion homeostasis. Sodium/bicarbonate cotransporter which plays an important role in regulating intracellular pH (PubMed : 18319254). Has been shown to act as a sodium/bicarbonate cotransporter in exchange for intracellular chloride (By similarity). Has also been shown to act as a sodium/biocarbonate cotransporter which does not couple net influx of bicarbonate to net efflux of chloride, with the observed chloride efflux being due to chloride self-exchange (PubMed : 18319254). Controls neuronal pH and may contribute to the secretion of cerebrospinal fluid (By similarity). Acting on presynaptic intracellular pH, it promotes GABA release, reduces the excitability of CA1 pyramidal neurons, and modulates short-term synaptic plasticity (By similarity).
