SLC4A8 often referred to as NBC or the sodium bicarbonate cotransporter is an integral membrane protein with a mass of around 129 kDa. Mechanically this protein mediates the transport of sodium and bicarbonate ions across cell membranes. It operates as an electroneutral cotransporter and is predominantly located in the plasma membranes of renal and neural cells. Expression of SLC4A8 is prominent in the kidney and brain where it controls acid-base balance and ionic gradients.
SLC4A8/NBC plays an essential role in maintaining intracellular pH and volume contributing to the acid-base homeostasis in the body. It does not function as part of a larger protein complex. This activity supports various cell functions including cell metabolism and neuronal activity. In kidney cells it assists in reabsorbing bicarbonate from the renal tubules which is important for the body’s pH regulation.
SLC4A8 participates in two important biological processes: the acid-base balance and bicarbonate reabsorption pathways. These pathways include the modulation of pH levels in renal and neural tissues. It connects directly with the NHE3 protein in the kidney which also takes part in sodium and proton exchange jointly influencing sodium reabsorption and extracellular fluid volume.
SLC4A8/NBC has associations with conditions like renal tubular acidosis and epilepsy. Dysfunctional expression or mutations in the SLC4A8 gene can lead to improper bicarbonate handling causing renal tubular acidosis which affects kidney function. In neurological disorders such as epilepsy altered ion transport through this protein can disrupt neuronal activity contributing to seizure activity. The protein interacts with GABAA receptors in the brain affecting inhibitory signaling which can be connected to epilepsy.
