Supplementary information

SLC6A15 also known as sodium-dependent amino acid transporter B(0)AT2 is a protein involved in the transport of amino acids across cellular membranes in a sodium-dependent manner. This protein's mass is approximately 73 kDa. It is expressed mostly in the brain kidney and small intestine where it plays an important role in maintaining amino acid homeostasis within cells.
Biological function summary

The transporter plays a significant role in neurotransmitter regulation by facilitating the uptake of neutral amino acids which serve as precursors for neurotransmitter synthesis. It is not known to be part of a larger complex which allows it functional specificity in the transport processes it mediates. The protein's activity impacts neurological functions and metabolic processes through its influence on amino acid availability.

Pathways

SLC6A15 significantly contributes to amino acid transport pathways particularly those critical for the central nervous system's function. This transporter interacts closely with proteins such as B0AT1 which also participates in similar transport activities. The pathways that incorporate SLC6A15 contribute to maintaining the balance of amino acids in the brain which are necessary for cognitive processes and metabolism.

SLC6A15's dysregulation has been associated with major depressive disorder and obesity. This link may be due to its vital role in amino acid supply for neurotransmitter production and metabolic regulation. Proteins such as serotonin transporter (SERT) might be affected due to alterations in amino acid availability impacting mood and metabolic balance. Understanding this protein's function in these conditions could offer insights into new therapeutic strategies.

Target data

Functions as a sodium-dependent neutral amino acid transporter. Exhibits preference for the branched-chain amino acids, particularly leucine, valine and isoleucine and methionine. Can also transport low-affinity substrates such as alanine, phenylalanine, glutamine and pipecolic acid. Mediates the saturable, pH-sensitive and electrogenic cotransport of proline and sodium ions with a stoichiometry of 1 : 1. May have a role as transporter for neurotransmitter precursors into neurons. In contrast to other members of the neurotransmitter transporter family, does not appear to be chloride-dependent.
