SLC7A7 also known as LAT1 or Lysinuric Protein Intolerance Protein is a component of the heterodimeric amino acid transporter complex. It weighs approximately 60 kDa. This protein functions mechanically by being part of a sodium-independent transporter system that facilitates the cellular import and export of essential amino acids particularly lysine arginine and ornithine. SLC7A7 mainly exhibits its expression in epithelial cells of the kidney and intestine as well as in the placenta and leukocytes reflecting its role in amino acid transport critical for cellular metabolism.
Biological function summary

This transporter protein plays an important role in balancing intracellular and extracellular concentrations of specific amino acids which are fundamental building blocks for protein synthesis and precursors for various metabolic processes. SLC7A7 forms a complex with a partner protein SLC3A2 to form a functional transporter. This complex localizes in cell membranes and actively participates in maintaining amino acid homeostasis which is essential for cell growth tissue repair and immune function.

Pathways

SLC7A7 is integral in the amino acid transport and metabolism pathways. It contributes to the mammalian target of rapamycin (mTOR) signaling pathway which regulates cell growth based on nutrient availability including amino acids. In this pathway it interacts with related proteins such as mTOR enabling cellular responses to nutrient levels. Additionally the SLC7A7 transporter is involved in the urea cycle and nitric oxide synthesis through its impact on arginine transport linking it to these metabolic processes.

SLC7A7 is strongly associated with Lysinuric Protein Intolerance (LPI) a rare autosomal recessive disorder characterized by poor absorption and excretion of cationic amino acids leading to growth retardation and immune dysfunction. The deficiency in functioning SLC7A7 disrupts normal amino acid transport which affects nutritional status and overall health. SLC7A7's involvement is also noted in systemic disorders related to mTOR pathway dysregulation linking it to conditions like certain metabolic syndromes. In the context of these disorders its interaction with SLC3A2 remains a point of significant consideration.

Heterodimer with SLC3A2, that functions as an antiporter which operates as an efflux route by exporting cationic amino acids from inside the cells in exchange with neutral amino acids plus sodium ions and may participate in nitric oxide synthesis via the transport of L-arginine (PubMed : 10080182, PubMed : 10655553, PubMed : 14603368, PubMed : 15756301, PubMed : 15776427, PubMed : 17329401, PubMed : 9829974, PubMed : 9878049). Also mediates arginine transport in non-polarized cells, such as monocytes, and is essential for the correct function of these cells (PubMed : 15280038, PubMed : 31705628). The transport mechanism is electroneutral and operates with a stoichiometry of 1 : 1 (By similarity). In vitro, Na(+) and Li(+), but also H(+), are cotransported with the neutral amino acids (By similarity).
