Slingshot homolog 1 (SSH1) also known as SSH1L is a protein phosphatase with a molecular mass of approximately 133 kDa. SSH1 is expressed in many tissues with higher levels in the brain and lung. It specifically dephosphorylates and activates cofilin a protein involved in actin dynamics. SSH1 contains a phosphatase domain essential for its activity which targets serine/threonine residues on substrates.
Biological function summary

SSH1 plays a critical role in the regulation of actin filament dynamics. It is not part of a known complex but interacts with several proteins involved in cytoskeleton organization. SSH1 reactivates cofilin by dephosphorylation facilitating the disassembly of actin filaments which is vital for processes such as cell migration division and morphogenesis. The ability of SSH1 to regulate actin dynamics makes it important for maintaining cell shape and motility.

Pathways

SSH1 contributes significantly to the actin dynamics pathway influencing cell architecture and movement. It operates downstream of the Rho family of GTPases which modulate actin reorganization. SSH1 also interacts with LIM kinase (LIMK) through the cofilin pathway. LIMK phosphorylates and inactivates cofilin while SSH1 reverses this action establishing a feedback loop critical for actin turnover.

SSH1 has implications in cancer and neurodegenerative diseases. In cancer abnormalities in SSH1 function can lead to enhanced cell migration and metastasis partly through interactions with cofilin and other cytoskeletal regulators. In neurodegenerative diseases altered SSH1 activity affects neuronal cell shape and connectivity. Dysregulation of cofilin by SSH1 is linked to the progression of these conditions highlighting the potential targeting of SSH1-related pathways for therapeutic interventions.

Dephosphorylates and activates the actin binding/depolymerizing factor cofilin, which subsequently binds to actin filaments and stimulates their disassembly. Inhibitory phosphorylation of cofilin is mediated by LIMK1, which may also be dephosphorylated and inactivated by this protein.
