Specificity

This affinity purified antibody is directed against human SLIT-3 protein. The product was affinity purified from monospecific antiserum by immunoaffinity purification. A BLAST analysis was used to suggest reactivity with this protein from human, rat, dog, bovine and mouse based on 100% homology for the immunogen sequence. Expect cross reactivity with SLIT-3 from chimpanzee and chicken sources, as only a single amino acid residue change is found within the immunogen sequence (93% positive by BLAST). Cross reactivity with SLIT-3 homologues from other sources has not been determined.

Slit3 also known as Slit Homolog 3 is a secreted protein encoded by the SLIT3 gene. It belongs to the Slit family of proteins and has a mass of approximately 160 kDa. These proteins typically have a leucine-rich domain and immunoglobulin-like repeats. Slit3 is expressed in various tissues including the heart lung and kidney but shows highest expression in the fetal lung. It operates in the extracellular space where it interacts with cell surface receptors to relay information essential for cellular functions.
Biological function summary

Slit3 regulates cellular migration and axon guidance by binding to Roundabout (Robo) receptors. This interaction plays a critical role in neuronal development and vascularization processes. In addition Slit3 can influence endothelial cell behavior and is a part of the axon guidance complex. By contributing to angiogenesis and neural patterning Slit3 impacts both developmental and repair mechanisms within the organism.

Pathways

The Slit3 protein participates in the axon guidance and angiogenesis signaling pathways. Slit3 interacts with Robo receptors to mediate cellular repulsion and navigation of the growth cones influencing nervous system architecture. The protein is closely related to other Slit family members particularly Slit1 and Slit2 which share similar roles in regulating guidance cues and vascular formation ensuring proper tissue development and injury response.

Disruptive mutations or altered expression of Slit3 have been linked to congenital diaphragmatic hernias and tumorigenesis. Congenital diaphragmatic hernia a defect that affects the diaphragm development connects to abnormal Slit3-mediated signaling that impacts tissue differentiation. Additionally changes in Slit3 expression might contribute to cancer progression by affecting tumor angiogenesis; this interaction involves proteins such as Robo1 and Robo4 making them potential targets in disease modulation.
