Transcriptional modulator that plays a role in various cellular processes, including embryonic development, cell differentiation, and tissue homeostasis (PubMed:9335504). Upon BMP ligand binding to their receptors at the cell surface, is phosphorylated by activated type I BMP receptors (BMPRIs) and associates with SMAD4 to form a heteromeric complex which translocates into the nucleus acting as transcription factor (PubMed:33667543). In turn, the hetero-trimeric complex recognizes cis-regulatory elements containing Smad Binding Elements (SBEs) to modulate the outcome of the signaling network (PubMed:33667543). SMAD1/OAZ1/PSMB4 complex mediates the degradation of the CREBBP/EP300 repressor SNIP1. Positively regulates BMP4-induced expression of odontogenic development regulator MSX1 following IPO7-mediated nuclear import (By similarity).

SMAD 1/5/8/9 also known by the genes SMAD1 SMAD5 SMAD8 and SMAD9 are integral proteins in the transforming growth factor-beta (TGF-β) signaling pathway. These proteins function as transcriptional modulators once activated. The molecular weight of SMAD1 SMAD5 and SMAD8 ranges from approximately 60 to 65 kDa. These proteins are expressed in various tissues and are especially active in bone cartilage and the vascular system. They act by transmitting extracellular signals from TGF-β superfamily ligands to the nucleus leading to gene expression changes.
SMAD1/5/8/9 proteins play essential roles in processes like cellular proliferation differentiation and apoptosis. These proteins typically form complexes with other SMAD proteins such as SMAD4 upon activation by type I receptor kinases. The formation of these complexes is important for their translocation to the nucleus where they regulate target gene expression. This regulation is key in controlling pivotal developmental and homeostatic processes across different cell types.
SMAD1/5/8/9 proteins are central components of the bone morphogenetic protein (BMP) signaling pathway. They mediate the signal transduction from BMPs which are important for bone development and embryogenesis. These proteins interact with other signaling molecules such as BMP receptors and various co-SMADs like SMAD4 to relay and modulate signals effectively. Through these interactions SMAD1/5/8/9 function within a well-defined network ensuring precise cellular responses to BMP stimuli.
SMAD1/5/8/9 proteins show significant involvement in conditions like cancer and pulmonary arterial hypertension (PAH). Aberrant BMP signaling through dysfunctions in these SMAD proteins has been linked to tumorigenesis where abnormal cell proliferation is a hallmark. Additionally alterations in SMAD1/5/8/9 activity are associated with PAH a condition characterized by high blood pressure in the pulmonary arteries.
