Supplementary information

Small integral membrane protein 1 (SMIM1) also known as Vel blood group antigen is a component of erythrocyte membranes. This protein has a molecular mass of about 9 kDa. While its expression is most observed in red blood cells SMIM1 may also exhibit presence in other tissue types though less extensively studied. SMIM1 spans the membrane indicating its integral role as a transporter or a facilitator for intercellular communications.
Biological function summary

The small protein influences the surface characteristics of erythrocytes. It impacts cell membrane stability and arrangement of other proteins in the lipid bilayer. While not forming large complexes itself SMIM1 interacts with various membrane components to preserve membrane integrity. Its presence contributes to erythrocyte shape which is critical for optimal blood flow and oxygen transport.

Pathways

SMIM1 participates in pathways related to erythrocyte development and function. It relates closely to the erythroblastic island a niche important for red blood cell maturation. Through its interactions SMIM1 associates with proteins like band 3 and glycophorin which are involved in maintaining cell structure and transport mechanisms. These associations emphasize SMIM1's involvement in the orchestration of red blood cell stability and viability.

Aberrant SMIM1 expression or mutations link to transfusion-related complications and blood-group incompatibility. Vel-negative blood group individuals lack SMIM1 leading to transfusion challenges. SMIM1 mutations can relate to disorders like anemia where abnormal erythrocyte morphology impairs function. These conditions often involve other membrane proteins such as spectrin highlighting SMIM1's role within a network important for erythrocyte health.
