Small leucine rich protein 1 (SLRP1) also known as osteoglycin or mimecan is a small proteoglycan with a molecular mass of approximately 36 kDa. SLRP1 contains leucine-rich repeats which are important for its function. It is predominantly found in the extracellular matrix of connective tissues including cartilage skin and bone where it plays a part in matrix organization. The expression of this protein can vary between different types of tissues highlighting its diverse roles in various physiological environments.
Biological function summary

Small leucine rich proteins such as SLRP1 are involved in the regulation of collagen fibrillogenesis. They interact directly with collagen fibrils and influence fiber size and spacing. SLRP1 is also known to impact cellular proliferation and differentiation which is partly due to its interaction with various growth factor receptors. This protein does not commonly form part of large stable complexes but may transiently associate with other extracellular matrix components during tissue development and repair.

Pathways

Several studies link small leucine rich proteins including SLRP1 to the transforming growth factor-beta (TGF-β) signaling pathway. TGF-β is important for controlling cell growth and repair mechanisms and SLRP1 can modulate its activity by influencing its bioavailability and receptor interactions. Additionally SLRP1 may have roles in the Wnt signaling pathway connected through indirect mechanisms related to extracellular matrix stability further interacting with other structural proteins in extracellular environments.

Malfunction or altered expression of SLRP1 associates primarily with musculoskeletal disorders such as osteoarthritis. Changes in the extracellular matrix composition and collagen interaction can disrupt tissue integrity contributing to disease progression. SLRP1 also associates with certain cardiovascular diseases where alterations in extracellular matrix components affect vascular function. In some cases the protein interacts with matrix metalloproteinases which degrade matrix proteins and contribute to structural changes in diseased tissues.
