Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-SMC2 antibody (AB10399)

IHC image of ab10399 staining in normal human cerebral cortex formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab10399, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Supplementary information

SMC2 also known as Structural Maintenance of Chromosomes Protein 2 is a critical component of the Condensin complex with a mass of approximately 135 kDa. This protein primarily functions within the nucleus of the cell where it plays an important role in chromosome condensation during cell division. SMC2 belongs to the SMC family of proteins and is widely expressed in various tissues including highly proliferative ones like bone marrow and thymus. The protein structure includes coiled-coil and hinge regions that are vital for its function in maintaining chromosome structure.
Biological function summary

SMC2 plays an essential role in ensuring proper chromosome segregation during mitosis. It acts as part of the Condensin complex a multi-protein assembly that is also composed of SMC4 CAP-D2 and CAP-G among others. This complex is important for the restructuring of chromosomes into compact forms allowing them to be easily separated during cell division. SMC2 binds to DNA and aids in the resolution of sister chromatids facilitating the equal distribution of genetic material to daughter cells.

Pathways

SMC2 operates within the cell cycle control pathway where it is involved in mitotic processes. It interacts directly with other components of the cell cycle machinery including Cyclin-dependent kinase 1 (CDK1). SMC2 also plays a role in the DNA repair pathway partnering with proteins like ATM (Ataxia-Telangiectasia Mutated kinase) which senses DNA damage and coordinates repair processes. These pathways highlight the interconnected nature of SMC2's functions in maintaining genetic stability.

SMC2 has been linked to certain cancers due to its pivotal role in ensuring proper mitosis. Disruptions in its function can lead to aneuploidy and genomic instability common features of tumorigenesis. Furthermore SMC2's involvement in DNA repair pathways implies a connection to disorders related to DNA damage like Ataxia-Telangiectasia. This disorder is marked by increased sensitivity to DNA damage and proteins like ATM are key partners of SMC2 in these repair processes. Understanding these links provides insights into potential therapeutic targets for addressing such diseases.

Target data

Central component of the condensin complex, a complex required for conversion of interphase chromatin into mitotic-like condense chromosomes. The condensin complex probably introduces positive supercoils into relaxed DNA in the presence of type I topoisomerases and converts nicked DNA into positive knotted forms in the presence of type II topoisomerases.
