Core component of the SMC5-SMC6 complex, a complex involved in DNA double-strand breaks by homologous recombination. The complex may promote sister chromatid homologous recombination by recruiting the SMC1-SMC3 cohesin complex to double-strand breaks. The complex is required for telomere maintenance via recombination in ALT (alternative lengthening of telomeres) cell lines and mediates sumoylation of shelterin complex (telosome) components which is proposed to lead to shelterin complex disassembly in ALT-associated PML bodies (APBs). Required for recruitment of telomeres to PML nuclear bodies. SMC5-SMC6 complex may prevent transcription of episomal DNA, such as circular viral DNA genome (PubMed:26983541).

SMC6 also known as Structural Maintenance of Chromosomes 6 is a core component of the SMC5/6 complex. It has a molecular mass of about 128 kDa. This protein is expressed in various tissues indicating its widespread role in cellular functions. SMC6 is known for its role in DNA repair and proper chromosome segregation during cell division where it maintains genomic stability.
SMC6 operates as part of the SMC5/6 complex important for DNA repair processes. The complex is involved in Homologous Recombination Repair (HRR) of DNA double-strand breaks. It also contributes to resolving DNA replication stress. SMC6 ensures that cells sustain genomic integrity by supporting chromosomal dynamics during the cell cycle.
The SMC6 protein is significant in DNA damage response and cell cycle checkpoint pathways. It cooperates closely with related proteins like RAD51 and BRCA1. These interactions are essential in maintaining genomic stability aiding in DNA repair and safe replication processes.
Disruption in SMC6 functions relates to cancer and Fanconi anemia. Its role in maintaining DNA integrity associates it with tumor suppression where impaired SMC6 can lead to genomic instability seen in cancers. In Fanconi anemia faulty coordination with other proteins such as FANCD2 affects DNA repair making SMC6 an important target to better understand these conditions.
