The Smek1 protein also known as SMEK homolog 1 or Protein Phosphatase 4 regulatory subunit 3A (PP4R3A) is involved in the regulation of protein phosphorylation. It has a molecular weight of approximately 97 kDa. Smek1 is expressed in various tissues including the brain and liver suggesting it plays roles in multiple biological functions. It acts by directing phosphatase activity towards specific targets making it essential for several signaling pathways.
Biological function summary

Smek1 contributes to cellular processes through its role as part of the protein phosphatase 4 (PP4) complex. This complex modulates several signaling pathways by dephosphorylating specific substrates which influences cell cycle progression apoptosis and DNA repair. As a regulatory subunit Smek1 enhances the specificity of PP4 to target phosphorylated proteins thereby participating in controlling cellular homeostasis and response to stress.

Pathways

Smek1 has essential roles in the DNA damage response and Wnt signaling. In the DNA damage response Smek1 helps control the dephosphorylation of proteins involved in DNA repair such as those encoded by the RAD51 and ATM genes. Through the Wnt signaling pathway Smek1 affects the regulation of beta-catenin an important mediator in cell growth and differentiation. These pathways show how Smek1 integrates into larger cellular processes by directing enzyme activity in response to various signals.

Smek1 holds significance in neurological and cancer conditions. In particular its role in DNA repair connects it to neurodegenerative diseases where proper repair mechanisms are vital to prevent neuronal damage. Furthermore alterations in Smek1 function have implications in oncogenesis especially where atypical Wnt signaling contributes to tumor development. Smek1 interacts with proteins such as beta-catenin in cancer progression highlighting its potential as a target for therapeutic intervention.
