Immunoprecipitation - Anti-SNM1A antibody (AB226003)

SNM1A was immunoprecipitated from HEK-293T (human epithelial cell line from embryonic kidney transformed with large T antigen) whole cell lysate (1 mg for IP, 20% of IP loaded) with ab226003 at 6 μg/mg lysate. Western blot was performed from the immunoprecipitate using ab226003 at 1 μg/ml.

Supplementary information

SNM1A also known as three-prime repair exonuclease 1 (TREX1) is a DNA repair enzyme with exonuclease activity. It plays an important role in the removal of DNA lesions and is important for maintaining genomic stability. The protein weighs approximately 109 kDa and is expressed in various tissues with higher expression in the bone marrow and lymphoid tissues. SNM1A cleaves DNA substrates aiding in DNA repair processes.
Biological function summary

SNM1A participates in DNA damage response and repair. It functions as part of the DNA interstrand cross-link repair and processes stalled replication forks. SNM1A does not form part of a larger protein complex but works alongside other proteins like FANCD2 and FANCI. Its activity is essential for preventing DNA damage accumulation which may lead to cell death or cancerous growths.

Pathways

SNM1A operates in the Fanconi anemia pathway facilitating the repair of interstrand cross-links which are critical obstacles to DNA replication. It associates with components like the SLX4 complex and the FANCD2-FANCI complex helping resolve replication issues. Another important pathway includes the homologous recombination repair interconnecting SNM1A with RAD51 a protein important in the exchange of genetic material for accurate DNA repair.

SNM1A is linked to predispositions in certain cancers due to its role in maintaining genomic integrity. Dysfunction in SNM1A activity associates with increased cancer risk particularly leukemias where genomic instability is a prominent feature. Additionally its relationship with FANCD2 within the Fanconi anemia pathway correlates SNM1A with Fanconi anemia-related disorders which showcase increased fragility and cancer susceptibility.

Target data

May be required for DNA interstrand cross-link repair. Also required for checkpoint mediated cell cycle arrest in early prophase in response to mitotic spindle poisons. Possesses beta-lactamase activity, catalyzing the hydrolysis of penicillin G and nitrocefin (PubMed : 31434986). Exhibits no activity towards other beta-lactam antibiotic classes including cephalosporins (cefotaxime) and carbapenems (imipenem) (PubMed : 31434986).
