Supplementary information

'Solo' also known as the SQSTM1-like protein is a multi-functional protein with a mass of approximately 62 kDa. It is expressed abundantly in the cytoplasm of various cell types and tissues particularly in muscle and nerve tissues. Solo functions mechanistically by serving as a scaffold protein that interacts with other proteins to regulate cellular processes such as autophagy and signal transduction. Its expression level can vary depending on the cellular context indicating its role in dynamic cellular modulation.
Biological function summary

Solo operates within the context of cellular maintenance and response mechanisms. It acts as an important regulator in autophagic and ubiquitin-proteasome systems often but not always working in conjunction with other proteins to form complexes. Solo modulates the degradation of misfolded proteins and damaged organelles ensuring cellular homeostasis and stress response. Its interactions may influence cellular survival pathways emphasizing Solo's participation in maintaining cellular integrity and normal function.

Pathways

Solo's involvement spans important biological processes such as the autophagy-lysosome pathway and the mTOR signaling pathway. In the autophagy pathway Solo interacts with proteins like LC3 and p62 playing an important role in cargo recognition and transport to lysosomes for degradation. In the mTOR pathway it influences cell growth and proliferation by modulating nutrient sensing and energy balance. Solo's interactions within these pathways highlight its role in regulating cellular metabolism and stress responses.

Solo has been associated with neurodegenerative conditions and muscular dystrophies. In diseases like Alzheimer's Solo's dysregulation can affect protein aggregation and clearance linking it to proteins such as tau and amyloid-beta. In muscular dystrophies alterations in Solo expression and function might impact muscle cell health associating it with dystrophin and related proteins. Understanding Solo's roles in these diseases can provide insights into therapeutic targets and strategies for intervention.

Target data

May act as the primary docking protein directing membrane turnover and assembly of the transient receptor potential channels TRPC4 and TRPC5. Binds phospholipids such as phosphatidylinositol monophosphates, phosphatidylinositol diphosphates (PIP2s) and phosphatidic acid, but not less polar lipids including phosphatidylcholine, phosphatidylserine, and phosphatidylinositol. The binding to PIP2s is calcium dependent. Might be involved in the plasma membrane localization of CTNNB1.
