Function

Polyol dehydrogenase that catalyzes the reversible NAD(+)-dependent oxidation of various sugar alcohols. Is mostly active with D-sorbitol (D-glucitol), L-threitol, xylitol and ribitol as substrates, leading to the C2-oxidized products D-fructose, L-erythrulose, D-xylulose, and D-ribulose, respectively (PubMed:3365415). Is a key enzyme in the polyol pathway that interconverts glucose and fructose via sorbitol, which constitutes an important alternate route for glucose metabolism. The polyol pathway is believed to be involved in the etiology of diabetic complications, such as diabetic neuropathy and retinopathy, induced by hyperglycemia (PubMed:12962626, PubMed:25105142, PubMed:29966615). May play a role in sperm motility by using sorbitol as an alternative energy source for sperm motility (PubMed:16278369). May have a more general function in the metabolism of secondary alcohols since it also catalyzes the stereospecific oxidation of (2R,3R)-2,3-butanediol. To a lesser extent, can also oxidize L-arabinitol, galactitol and D-mannitol and glycerol in vitro. Oxidizes neither ethanol nor other primary alcohols. Cannot use NADP(+) as the electron acceptor (PubMed:3365415).

Activity summary

Sorbitol Dehydrogenase (SDH) also known as SORD is an enzyme that catalyzes the oxidation of sorbitol to fructose. This reaction involves the conversion of a secondary alcohol group to a ketone. SDH has a molecular mass of approximately 38 kDa. It is expressed in various human tissues most importantly in the liver kidneys and seminal vesicles. The enzyme requires NAD+ as a cofactor for its catalytic activity.

Biological function summary

SDH plays a part in the polyol pathway which is important for the metabolism of glucose into fructose. It functions alongside aldose reductase to regulate the conversion of glucose into sorbitol and then sorbitol into fructose. SDH acts independently without forming a complex with other proteins. The amount and activity of SDH can affect cellular osmotic balance and energy storage.

Pathways

SDH is involved in the polyol pathway and influences sorbitol levels. An excess of sorbitol can contribute to cellular damage especially in the context of diabetes. SDH’s role is linked to the exclusive conversion of sorbitol to fructose contrasting with aldose reductase which initiates sorbitol production from glucose. Both enzymes are key in maintaining the balance between extracellular and intracellular glucose.

Associated diseases and disorders

Alterations in SDH activity relate to diabetic complications like diabetic retinopathy and neuropathy. The accumulation of sorbitol due to insufficient SDH activity contributes to the development of these conditions. SDH's connection with aldose reductase is particularly notable here as both proteins impact the sorbitol levels that affect various tissues under hyperglycemic conditions.
