Immunocytochemistry/ Immunofluorescence - Anti-SorCS1 antibody (AB237618)

MCF7 (human breast adenocarcinoma cell line) cells stained for SorCS1 (green) using ab237618 at 1/100 dilution in ICC/IF, followed by Alexa Fluor 488-congugated Goat Anti-Rabbit IgG (H+L) secondary antibody.

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. Counterstained with DAPI.

Supplementary information

SorCS1 also known as Sortilin-related Vps10 domain containing receptor 1 is a multifunctional protein with a mass of approximately 130 kDa. The protein primarily operates as a sorting receptor within cells influencing protein trafficking. It can bind neurotrophins regulate their transport and manage degradation pathways. SorCS1 is expressed in various tissues including the central nervous system muscle and pancreas. Notably it functions on the cell surface but is also present in distinct intracellular compartments.
Biological function summary

This protein impacts neuronal development and glucose metabolism. SorCS1 belongs to the Vps10p domain receptor family which partakes in protein sorting processes. It is involved in amyloid-beta metabolism and modulates insulin receptor signaling. Through these interactions SorCS1 indirectly affects energy balance and brain function making it significant for neural cell maintenance and metabolic regulation.

Pathways

SorCS1 interacts with cellular mechanisms like insulin signaling and amyloid-beta processing. It modulates the insulin signaling pathway by interacting with the insulin receptor. SorCS1 also influences the amyloid precursor protein (APP) processing pathway impacting the production of amyloid-beta. These pathways link SorCS1 functionally to proteins like sortilin and low-density lipoprotein receptor-related protein 1 (LRP1) highlighting its role in intersecting signaling mechanisms.

Research indicates SorCS1's connection to diabetes and Alzheimer's disease. It can influence the development of type 2 diabetes by modulating insulin signaling pathways and impacting glucose homeostasis. SorCS1 affects the accumulation of amyloid-beta peptides an important factor in Alzheimer's disease progression. SorCS1’s relationships with proteins such as APP and LRP1 deepen its relevance to these conditions presenting it as a potential therapeutic target for intervention.
