Acts as a component of the essential kinetochore-associated NDC80 complex, which is required for chromosome segregation and spindle checkpoint activity (PubMed:14738735). Required for kinetochore integrity and the organization of stable microtubule binding sites in the outer plate of the kinetochore (PubMed:14738735). The NDC80 complex synergistically enhances the affinity of the SKA1 complex for microtubules and may allow the NDC80 complex to track depolymerizing microtubules (PubMed:23085020).

Activity summary

The SPC24 protein also known as Spindle Pole Body Component 24 weighs around 22 kDa. SPC24 is expressed in various tissues but it shows higher levels in dividing cells particularly during mitosis. Mechanically it forms an essential part of the NDC80 complex contributing to kinetochore-microtubule connections that are important for chromosome segregation during cell division. SPC24 interacts with microtubules through the NDC80 complex ensuring accurate chromosome alignment and stabilization.

Biological function summary

SPC24 plays a role in maintaining genomic stability and its function is highly dependent on the NDC80 complex. This complex is an integral component of the kinetochore a structure on chromosomes critical for equal distribution during cell division. By anchoring microtubules to kinetochores SPC24 helps prevent aneuploidy which can arise from improper chromosomal segregation. The presence of SPC24 supports robust kinetochore-microtubule attachments which are important for precise mitotic progression and cellular proliferation.

Pathways

SPC24 is involved in the cell cycle especially during mitosis. In the mitotic pathway it assists in the regulation of spindle checkpoint signaling ensuring each daughter cell receives the correct number of chromosomes. This ensures proper mitotic checkpoint control. SPC24 interfaces with other proteins such as SPC25 within the NDC80 complex and interacts with signaling pathways involving kinases like Aurora B which monitors and corrects kinetochore-microtubule attachments.

Associated diseases and disorders

SPC24 plays a role in cancer progression notably in cases where chromosomal instability is high such as in colorectal cancer. Aberrations in SPC24 function or expression can lead to improper kinetochore tension and chromosomal missegregation worsening this instability. SPC24 can relate to the enhancement of aggressive cancer phenotypes acting in conjunction with proteins like Mis12 which also contributes to kinetochore function. Targeting SPC24 could offer pathways for therapeutic strategies in treating cancers marked by mitotic errors.
