Supplementary information

This supplementary information is collated from multiple sources and compiled automatically.

SPCS2 also called SPC25 is a component of the signal peptidase complex involved in cleaving signal peptides from nascent proteins during translocation into the endoplasmic reticulum. This protein has a molecular mass of approximately 27 kDa. SPCS2 is expressed in various tissues with notable levels in the liver pancreas and kidney. This broad expression suggests its importance across multiple physiological systems.
Biological function summary

The function of SPCS2/SPC25 relates to its role in the signal peptidase complex. It participates in the maturation of proteins by processing them for proper folding and export within the secretory pathway. As part of this multimeric complex in the endoplasmic reticulum membrane SPCS2 coordinates with other components like SEC11 to efficiently cleave signal peptides. Its function is critical for the regulation of protein synthesis and secretion processes.

Pathways

SPCS2 functions within the secretory pathways particularly in protein translocation into the endoplasmic reticulum. Its involvement in the ER-associated degradation pathway highlights its role in maintaining protein homeostasis. Related proteins such as SEC61 and ERAB are key associates in these pathways working together to facilitate the translocation and processing of nascent proteins entering the ER.

SPCS2 is linked with pancreatic diseases and certain liver dysfunctions due to its role in protein processing and secretion. Aberrant functioning of SPCS2 can disrupt normal protein folding and maturation potentially leading to conditions like pancreatitis. The protein's link to SEC11 in the protein processing pathways highlights shared dysfunction pathways that can influence disease severity and progression.

Target data

Component of the signal peptidase complex (SPC) which catalyzes the cleavage of N-terminal signal sequences from nascent proteins as they are translocated into the lumen of the endoplasmic reticulum (PubMed : 34388369). Enhances the enzymatic activity of SPC and facilitates the interactions between different components of the translocation site (By similarity).
