Western blot - Anti-Spectrin beta-3 antibody [RP23040210] (AB308134)
Western blot analysis was performed on whole cell extracts (30 µg lysate) of HeLa (Lane 1), HT-29 (Lane 2), HCT 116 (Lane 3), Caco-2 (Lane 4), MCF7 (Lane 5), T47D (Lane 6), THP-1 (Lane 7) and KARPAS 299 (Lane 8). The blots were probed with Anti-Spectrin Beta 3 Recombinant Rabbit Polyclonal Antibody (ab308134, 1-2 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate (0.4 µg/mL, 1 : 2500 dilution). A 268 kDa band corresponding to Spectrin Beta 3 was observed across cell lines tested. Known quantity of protein samples were electrophoresed using a 4-12% Bis-Tris gel, electrophoresis system and pre-stained protein standard. Resolved proteins were then transferred onto a nitrocellulose membrane with an overnight wet transfer system. The membrane was probed with the relevant primary and secondary Antibody following blocking with 5% skimmed milk. Chemiluminescent detection was performed (ECL).
