Activity summary

Spermidine also known as SPD is a polyamine with a molecular mass of approximately 145.25 g/mol. It originates from the decarboxylation product of putrescine and is synthesized in the cytoplasm. Spermidine expresses ubiquitously in both prokaryotic and eukaryotic cells. Its mechanical role involves stabilizing the structure of the ribosome and enhancing nucleic acid functions. Through its interaction with RNA it influences cellular functions like growth and proliferation.

Biological function summary

This polyamine plays an important role in cellular processes such as autophagy where it acts as an important regulator. Spermidine is not part of a larger protein complex but it interacts directly with other cellular components to modulate cellular homeostasis and stress resistance. Additionally spermidine is involved in processes like telomerase activity and apoptosis proving its significance in cell maintenance and longevity.

Pathways

Spermidine significantly contributes to the polyamine biosynthesis and catabolism pathways. It is involved in the regulation of the mechanistic target of rapamycin (mTOR) pathway which is important for controlling cellular growth and metabolism. Spermidine functions in connection with proteins like ornithine decarboxylase (ODC) and S-adenosylmethionine decarboxylase (SAMDC) which are pivotal for the polyamine synthesis pathway and interconnected processes.

Associated diseases and disorders

Spermidine shows relevance to conditions such as neurodegenerative disorders and certain cancers. Its ability to modulate autophagy suggests its importance in mitigating the progression of neurodegenerative diseases like Alzheimer's. Additionally spermidine's role in cell growth regulation links it to cancer as dysregulated polyamine metabolism often correlates with tumorigenesis. In these conditions spermidine interacts with proteins such as Beclin 1 in autophagy and MYC in cell growth regulation highlighting its therapeutic potential in managing these diseases.

    ab7318 staining Spermidine in rat lungs by Immunohistochemistry (Formalin/ PFA fixed paraffin-embedded tissue sections). Tissue underwent fixation in 4% paraformaldehyde overnight and embedded in paraffin, heat mediate antigen retrieval was performed in target retrieval solution and sections were immersed in 3% hydrogen peroxide for 5 min to deplete the endogenous peroxidase activity. Sections were incubated in 1% bovine serum albumin in PBS for 30 min followed by primary antibody at room temperature for 2 h. A primary antibody Enhancer and HRP polymer were used according to the manufacturer's instructions. Left image show staining with ab7318 while the right one show staining with same procedure without primary antibody.
