Immunocytochemistry/ Immunofluorescence - Anti-Spi2A antibody [MoFo 29.2] (AB36772)
Immunofluorescent analysis of Spi2A (green) showing staining in the cytoplasm and nucleus of Jurkat cells (right) compared to a negative control without primary antibody (left). Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with ab36772 in 3% BSA-PBS at a dilution of 1 : 20 and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight®-conjugated secondary antibody in PBS at room temperature in the dark. Nuclei were stained with Hoechst or DAPI (blue). Images were taken at a magnification of 60x.

Biological function summary
Spi2A acts by inhibiting the activity of granzyme B a serine protease released by cytotoxic T cells and NK cells. Through its inhibition of granzyme B Spi2A protects target cells from apoptosis induced by the immune system. It is not part of a larger protein complex but functions as a standalone inhibitor. This activity allows immune cells to regulate cell death maintaining homeostasis and preventing excessive tissue damage during immune responses.
Pathways
Spi2A contributes significantly within immune regulatory pathways by influencing the apoptosis pathway. It primarily affects pathways associated with the immune response specifically in the management of cell-mediated cytotoxicity. Spi2A also interacts with other proteins involved in immune regulation such as caspases which play critical roles in apoptosis.
