Immunofluorescent analysis of Spi2A (green) showing staining in the cytoplasm and nucleus of Jurkat cells (right) compared to a negative control without primary antibody (left). Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with ab36772 in 3% BSA-PBS at a dilution of 1 : 20 and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight®-conjugated secondary antibody in PBS at room temperature in the dark. Nuclei were stained with Hoechst or DAPI (blue). Images were taken at a magnification of 60x.

Spi2A also known as Serpinb9 is a serine protease inhibitor that plays a mechanical role in regulating protease activity. Its molecular weight is approximately 42 kDa. Scientists find Spi2A expression in the cytoplasm of various immune cells including T lymphocytes and natural killer (NK) cells. It is expressed in specific tissues such as the spleen and lymph nodes which indicates its role in immune function.
Biological function summary

Spi2A acts by inhibiting the activity of granzyme B a serine protease released by cytotoxic T cells and NK cells. Through its inhibition of granzyme B Spi2A protects target cells from apoptosis induced by the immune system. It is not part of a larger protein complex but functions as a standalone inhibitor. This activity allows immune cells to regulate cell death maintaining homeostasis and preventing excessive tissue damage during immune responses.

Pathways

Spi2A contributes significantly within immune regulatory pathways by influencing the apoptosis pathway. It primarily affects pathways associated with the immune response specifically in the management of cell-mediated cytotoxicity. Spi2A also interacts with other proteins involved in immune regulation such as caspases which play critical roles in apoptosis.

The dysregulation of Spi2A has been linked to autoimmune diseases and certain cancers. Aberrant Spi2A function can contribute to autoimmune conditions by altering normal immune cell apoptosis leading to inappropriate cell survival. Additionally Spi2A interactions with proteins like granzyme B can affect the progression of certain cancers as cancer cells may express higher levels of Spi2A to evade immune-mediated apoptosis. Understanding Spi2A's regulation is therefore essential in these pathological conditions.

Serine and cysteine protease inhibitor. Can inhibit lysosomal papain-like proteases including the cathepsins B, G, H, K, L and V. Ineffective against elastase, granzyme A, granzyme B, or caspases 3, 8 or 9. Inhibition of cytoplasmic cathepsin B following release from the lysosome may protect cells from apoptosis. This may facilitate the survival of progenitor T-cells and the subsequent development of long term memory CD8 T-cells.
