Supplementary information

This supplementary information is collated from multiple sources and compiled automatically.

Spinophilin also known as Neurabin 2 is a protein with a molecular weight of approximately 120 kDa. This protein is mostly found in the central nervous system where it associates with dendritic spines of neurons. Spinophilin contains an actin-binding domain a PDZ domain and a coiled-coil domain which allows it to interact with various proteins involved in synaptic function. Its distribution is not limited to the brain as it has also been identified in other tissues like the liver and kidneys.
Biological function summary

Spinophilin functions in the regulation of synaptic signaling and structural plasticity contributing to neural communication. It often forms part of signaling complexes binding to protein phosphatase 1 (PP1) to modulate dephosphorylation activities. Spinophilin influences the organization and dynamics of actin filaments affecting the stability and morphology of dendritic spines which are critical sites for synaptic transmission and plasticity. Through these interactions Spinophilin plays an important role in learning and memory processes.

Pathways

Spinophilin interacts with systems involved in synaptic signaling and cytoskeletal organization. It impacts the RhoA/ROCK pathway which is essential for actin filament dynamics by modulating PP1 activity. Additionally Spinophilin indirectly affects the mTOR pathway which is involved in protein synthesis and cell growth. It relates to proteins such as RhoA and mTOR through its role in maintaining the balance of phosphorylation states within the cell.

Spinophilin's association with synaptic signaling links it to neurodegenerative diseases such as Alzheimer's disease where synaptic dysfunction is a characteristic feature. Furthermore Spinophilin's involvement in spine morphology suggests a role in psychiatric disorders like schizophrenia where altered spine density and structure are observed. In these diseases Spinophilin connects with proteins like tau in Alzheimer's and dopamine receptors in schizophrenia highlighting its potential as a therapeutic target.

Product protocols

For this product, it's our understanding that no specific protocols are required. You can visit:

Target data

Seems to act as a scaffold protein in multiple signaling pathways. Modulates excitatory synaptic transmission and dendritic spine morphology. Binds to actin filaments (F-actin) and shows cross-linking activity. Binds along the sides of the F-actin. May play an important role in linking the actin cytoskeleton to the plasma membrane at the synaptic junction. Believed to target protein phosphatase 1/PP1 to dendritic spines, which are rich in F-actin, and regulates its specificity toward ion channels and other substrates, such as AMPA-type and NMDA-type glutamate receptors. Plays a role in regulation of G-protein coupled receptor signaling, including dopamine D2 receptors and alpha-adrenergic receptors. May establish a signaling complex for dopaminergic neurotransmission through D2 receptors by linking receptors downstream signaling molecules and the actin cytoskeleton. Binds to ADRA1B and RGS2 and mediates regulation of ADRA1B signaling. May confer to Rac signaling specificity by binding to both, RacGEFs and Rac effector proteins. Probably regulates p70 S6 kinase activity by forming a complex with TIAM1 (By similarity). Required for hepatocyte growth factor (HGF)-induced cell migration.
