Plays a critical role in the phospholipid salvage pathway from lysosomes to the cytosol (PubMed:36161949, PubMed:37075117). Mediates the rate-limiting, proton-dependent, lysosomal efflux of lysophospholipids, which can then be reacylated by acyltransferases in the endoplasmic reticulum to form phospholipids (PubMed:36161949, PubMed:37075117). Selective for zwitterionic headgroups such as lysophosphatidylcholine (LPC) and lysophosphatidylethanolamine (LPE), can also transport lysophosphatidylglycerol (LPG), but not other anionic lysophospholipids, sphingosine, nor sphingomyelin (PubMed:36161949). Transports lysophospholipids with saturated, monounsaturated, and polyunsaturated fatty acids, such as 1-hexadecanoyl-sn-glycero-3-phosphocholine, 1-(9Z-octadecenoyl)-sn-glycero-3-phosphocholine and 1-(4Z,7Z,10Z,13Z,16Z,19Z-docosahexaenoyl)-sn-glycero-3-phosphocholine, respectively (PubMed:36161949, PubMed:37075117). Can also transport lysoplasmalogen (LPC with a fatty alcohol) such as 1-(1Z-hexadecenyl)-sn-glycero-3-phosphocholine (PubMed:36161949). Lysosomal LPC could function as intracellular signaling messenger (PubMed:37075117). Essential player in lysosomal homeostasis (PubMed:36161949). Crucial for cell survival under conditions of nutrient limitation (PubMed:37075117). May be involved in necrotic or autophagic cell death (PubMed:12815463).

SPNS1 also known as Spinster homolog 1 is a transporter protein with a molecular weight of approximately 39 kDa. This protein is mainly found in tissues such as placenta brain lung and kidney. Mechanically SPNS1 functions as a transporter that facilitates the movement of sphingosine-1-phosphate (S1P) across the cellular membranes. This transport activity plays an essential role in regulating S1P levels within and outside cells impacting various cellular processes.
SPNS1 serves as a pivotal regulator in lipid signaling and cell migration. It is part of a system that balances the synthesis and degradation of S1P influencing numerous cellular functions including immune cell trafficking and vascular development. SPNS1’s activity is integral to maintaining the cellular gradient of S1P necessary for directing cellular responses in physiological contexts like immune response and cellular development. Its function links to several cellular outcomes making it a significant player in cellular communication.
SPNS1 significantly contributes to the sphingolipid signaling pathway and the vascular endothelial growth factor (VEGF) pathway. Within the sphingolipid signaling pathway SPNS1 works closely with sphingosine kinases which phosphorylate sphingosine to S1P and S1P receptors that mediate downstream signaling. In the VEGF pathway SPNS1 indirectly influences angiogenesis through lipid signaling mediators. ICC/IF image of ab59971 stained SHSY5Y cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab59971, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
