The Spry-4 protein also known as Sprouty 4 is an inhibitor of receptor tyrosine kinase signaling pathways. It weighs approximately 35 kDa and plays a role in modulating fibroblast growth factor (FGF) and epidermal growth factor (EGF) signaling. Spry-4 is expressed in various tissues including the brain lungs and kidneys. Its expression varies during different developmental stages and under different physiological conditions.
Biological function summary

Sprouty family proteins such as Spry-4 negatively regulate cell growth differentiation and angiogenesis. This protein functions as part of a negative feedback loop in growth factor-mediated signaling. By modulating signaling Spry-4 helps maintain balanced cellular responses ensuring cells do not undergo uncontrolled proliferation or differentiation. Spry-4's regulatory role is important in tissue development and repair.

Pathways

Spry-4 plays a significant role in the MAPK/ERK and PI3K/AKT pathways. In these pathways Spry-4 interacts with other proteins like RAF1 and Erk. Its inhibitory effects prevent excessive activation of these pathways which are essential for processes like cell division and survival. Spry-4's ability to modulate these pathways determines how cells respond to external stimuli maintaining cellular homeostasis.

Spry-4's dysregulation has been linked to cancer and congenital disorders. For example altered expression of Spry-4 can lead to tumorigenesis since the balance in growth factor signaling is disrupted. In cancer Spry-4 often interacts with other regulatory proteins such as SOS1 which further modulates signaling pathways important for tumor growth. In congenital disorders mutations in Spry-4 or related pathways affect normal tissue development leading to developmental anomalies.
