Supplementary information

Single-stranded DNA-binding protein 2 (SSBP2) also known as hSSB2 is a protein with a molecular mass of approximately 38 kDa. This protein plays a role in binding to single-stranded DNA. It is highly expressed in hematopoietic tissues including bone marrow and blood cells. SSBP2 also shows distinct expression in multiple human tissues highlighting its importance across different cellular environments. Mechanistically SSBP2 stabilizes single-stranded DNA by preventing the formation of secondary structures which is critical during DNA replication and repair.
Biological function summary

SSBP2 functions as a part of the multi-protein complex important for DNA metabolism and maintenance of genomic stability. It plays a role in the transcriptional activation of several tumor suppressor genes by interacting with other single-stranded DNA-binding partners. The protein influences cell cycle regulation by modulating the expression of genes involved in cell division and proliferation. This biological action affirms SSBP2's contribution to maintaining normal cellular functions and preventing abnormal cellular growth.

Pathways

The involvement of SSBP2 in cell cycle regulation and DNA repair underlines its participation in pathways like p53 signaling and homologous recombination. SSBP2 connects with proteins like RPA which is central to DNA metabolism to ensure accurate DNA replication and repair. These interactions place SSBP2 in a critical position to respond to and repair DNA damage thereby maintaining cellular integrity and genomic stability.

SSBP2 has relevance in leukemia and breast cancer. Its reduced expression or inactivity correlates with the progression of these diseases. In leukemia SSBP2's interaction with tumor suppressor proteins such as pRB is significant. In breast cancer its dysregulation can lead to unchecked cell proliferation. Understanding these relationships provides insight into SSBP2's potential as a biomarker and therapeutic target in cancer research.
