Ssu72 also known as SSU72 homolog RNA polymerase II CTD phosphatase is an enzyme with a mass of approximately 24 kDa. It functions as a specific phosphatase that targets the C-terminal domain (CTD) of RNA polymerase II. Ssu72 performs its mechanical role by removing phosphate groups from serine 5 residues within the heptad repeats of the CTD which is important for the transcription cycle. The expression of Ssu72 occurs in the nucleus where its activity aligns with its role in transcription regulation.
The enzymatic function of Ssu72 contributes to gene expression regulation. It plays a significant role during the transition between the transcription initiation and elongation phases. Ssu72 is an integral part of the transcription machinery and is known to associate with the cleavage and polyadenylation complex highlighting its involvement in the pre-mRNA processing. Additionally Ssu72's phosphatase activity helps regulate the interaction between RNA polymerase II and multiple transcription factors and complexes emphasizing its role in maintaining efficient transcription and RNA processing dynamics.
Ssu72 functions within the transcription regulation pathway where it interacts with other critical transcription-related proteins such as the Symplekin and TFIIB. Its involvement in the regulation of RNA polymerase II activity connects it to RNA processing pathways including mRNA capping splicing and termination processes. These interactions facilitate the precise control of gene expression by modulating transcription elongation and mRNA processing events ensuring coordinated transcriptional responses.
The dysfunction of Ssu72 can lead to transcriptional dysregulation contributing to cancer and other transcription-associated disorders. Disruptions in its phosphatase activity may have downstream effects on RNA polymerase II and the associated transcription regulation processes. Ssu72's connection to cancer is further supported by its interaction with proteins like PinX1 a negative regulator of telomerase where altered expression or activity could influence cellular proliferation and stability potentially driving oncogenesis.
