STAC3 also known as SH3 and cysteine-rich domain 3 is a protein with a mass of approximately 35 kDa. It performs an important mechanical role in the process of excitation-contraction coupling in skeletal muscle cells. STAC3 is expressed mainly in skeletal muscle tissues where it joins other essential proteins to ensure proper muscle function. By interacting with other key proteins in the muscle STAC3 assists in the regulation of calcium ion release which is vital for muscle contraction and performance.
Biological function summary

STAC3 functions as part of a protein complex that includes the dihydropyridine receptor (DHPR) and the ryanodine receptor (RYR1) in skeletal muscles. It serves as a linking molecule within this complex facilitating communication between these receptors. This linkage is significant for maintaining the efficiency of calcium release during muscle activation. Without proper communication aided by STAC3 the calcium signaling in muscle cells becomes impaired which can affect muscle contraction.

Pathways

STAC3 plays an important role in the ECC (excitation-contraction coupling) pathway. This pathway is critical in converting an electrical signal to a mechanical response in muscle cells. It interacts specifically with DHPR and RYR1 which are central components in this signaling process. Defects in STAC3 link to congenital myopathies specifically Native American myopathy (NAM). Required for normal Ca(2+) release from the sarcplasmic reticulum, which ultimately leads to muscle contraction. Probably functions via its effects on muscle calcium channels (PubMed : 23736855, PubMed : 29078335). Increases CACNA1S channel activity, in addition to its role in enhancing the expression of CACNA1S at the cell membrane. Has a redundant role in promoting the expression of the calcium channel CACNA1S at the cell membrane (By similarity). Slows down the inactivation rate of the calcium channel CACNA1C (PubMed : 29078335).
