Signal transducer and activator of transcription that mediates signaling by type I interferons (IFN-alpha and IFN-beta). Following type I IFN binding to cell surface receptors, Jak kinases (TYK2 and JAK1) are activated, leading to tyrosine phosphorylation of STAT1 and STAT2. The phosphorylated STATs dimerize, associate with IRF9/ISGF3G to form a complex termed ISGF3 transcription factor, that enters the nucleus. ISGF3 binds to the IFN stimulated response element (ISRE) to activate the transcription of interferon stimulated genes, which drive the cell in an antiviral state (PubMed:9020188, PubMed:23391734). In addition, has also a negative feedback regulatory role in the type I interferon signaling by recruiting USP18 to the type I IFN receptor subunit IFNAR2 thereby mitigating the response to type I IFNs (PubMed:28165510). Acts as a regulator of mitochondrial fission by modulating the phosphorylation of DNM1L at 'Ser-616' and 'Ser-637' which activate and inactivate the GTPase activity of DNM1L respectively (PubMed:26122121, PubMed:23391734, PubMed:9020188).

Sandwich ELISA of Human STAT2 Antibody Pair - BSA and Azide free ab322476 with the capture antibody Anti-STAT2 antibody [EPR29255-538] - BSA and Azide free (Capture) ab322477 dilution at 2 ug/mL and detector antibody ab322478 dilution at 0.5 ug/mL.
Interpolated concentration of native STAT2 was measured at different sample concentrations. Undiluted samples are as follows: Hela cell extract (500 µg/mL) and K562 cell extract (1000 µg/mL). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in antibody diluent CP2 (Antibody Diluent CP2 ab319981). Data represented here are indicative of assay working using example of biological sample. However, we do not guarantee same sensitivity level due to biological variation.
Interpolated concentrations of native STAT2 in human control wild type HeLa (human cervical adenocarcinoma epithelial cell) and STAT2 knockout HeLa cell based on 500 µg/mL extract loads. The concentrations of STAT2 were measured in duplicate and interpolated from the STAT2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean STAT2 concentration was determined to be 105.9 pg/mL in wild type HeLa extract (Human wild-type HeLa cell line Human wild-type HeLa cell line ab255928) and undetectable in STAT2 knockout HeLa extract (Human STAT2 knockout HeLa cell line Human STAT2 knockout HeLa cell line ab261819).
