Activity summary

Signal Transducer and Activator of Transcription 5 commonly known as STAT5 is a member of the STAT protein family. It weighs approximately 94 kDa. STAT5 usually exists in two closely related forms STAT5a and STAT5b with both playing similar roles but having slightly different activity. This protein is widely expressed found in cells like lymphocytes epithelial cells and endothelial cells indicating its importance in various tissues.

Biological function summary

STAT5 acts as a transcription factor that modulates gene expression by binding to specific DNA sequences. It often forms part of a larger complex when activated. This complex relocation to the nucleus triggers transcriptional activities essential for cell growth differentiation and survival. As it interacts with other proteins STAT5 plays an important role in cytokine signaling particularly in response to interleukins and growth hormones.

Pathways

STAT5 is important in the JAK-STAT signaling pathway. This pathway is activated by cytokines and growth factors leading to STAT5 phosphorylation and its subsequent translocation into the nucleus. The interaction with Janus kinases (JAKs) facilitates this process. STAT5 also plays a significant role in the MAPK pathway linking it to various cellular responses and functions.

Associated diseases and disorders

STAT5 has a strong association with oncological conditions like leukemia and lymphomas. Aberrant activation or dysregulation of STAT5 can lead to uncontrolled cell proliferation and survival contributing to cancer. Furthermore STAT5 is linked with autoimmune diseases with its malfunction affecting immune response and leading to disorders. In these conditions proteins like BCL-XL often interact with STAT5 influencing cell survival pathways.

    Z- and S- Score: The Z-score represents the strength of a signal that a monoclonal antibody (Monoclonal Antibody) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProtTM array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If targets on HuProtTM are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-score. S-score therefore represents the relative target specificity of a Monoclonal Antibody to its intended target. A Monoclonal Antibody is considered to specific to its intended target, if the Monoclonal Antibody has an S-score of at least 2.5. For example, if a Monoclonal Antibody binds to protein X with a Z-score of 43 and to protein Y with a Z-score of 14, then the S-score for the binding of that Monoclonal Antibody to protein X is equal to 29.
