STEAP3 also known as Six-Transmembrane Epithelial Antigen of Prostate 3 or TSAP6 acts as a metalloreductase. It is involved in the reduction of metal ions particularly iron. The protein facilitates the conversion of ferric to ferrous iron which is essential for cellular uptake of iron through divalent metal transporters. STEAP3 with a molecular weight of approximately 50 kDa is mainly expressed in prostate tissue but also found in other tissues like the liver and testis.
Biological function summary

STEAP3 plays an important role in iron homeostasis by contributing to iron metabolism and mobilization. The protein represents a component of the multiprotein complex in endosomes where it participates alongside proteins such as ferrireductase and divalent metal transporter 1 (DMT1). These interactions are important for maintaining proper cellular iron levels and preventing iron deficiency or overload.

Pathways

Iron metabolism and recycling are heavily influenced by the activities of STEAP3. The protein is important within the endosomal ferrireductase pathway where it collaborates closely with DMT1 and transferrin receptor proteins to mediate iron uptake and release. Additionally STEAP3 participates in the heme biosynthesis pathway indirectly by ensuring adequate availability of iron which is central to heme production and function.

Researchers have observed associations between STEAP3 dysregulation and conditions like anemia and cancer. Deficient STEAP3 activity can contribute to iron-related disorders such as anemia linked via its relation to the transferrin receptor which mediates iron uptake in cells. Conversely overexpression of STEAP3 has implications in cancer potentially linked to its interactions with p53 a well-known tumor suppressor protein indicating a role in cancer cell iron metabolism and growth.
