STIL also known as SIL (SCL/TAL1 Interrupting Locus) is a protein involved mechanically in cellular processes that are important for cell division. STIL has a molecular mass of approximately 150 kDa. This protein localizes primarily in the nucleus where it is abundantly expressed in actively dividing cells. It plays a direct role in maintaining proper cell cycle progression especially during mitosis.
Biological function summary

The STIL protein plays a role in regulating centrosome duplication ensuring accurate chromosome segregation. STIL forms part of a complex with other proteins like SAS-6 which is important in forming the mitotic spindle apparatus. This action is essential to maintaining the structural integrity of centrosomes which are key for correct cell division.

Pathways

The STIL protein integrates into cell cycle regulation and the centrosome cycle. STIL interacts with proteins such as PLK4 and CEP135 within the centrosome duplication pathway. This interaction is important for the initiation of procentriole biogenesis aligning with the G1 phase of the cell cycle.

Misregulation of the STIL protein associates strongly with cancer and microcephaly. For instance overexpression of STIL links to various cancers as it can lead to abnormal cell proliferation. Additionally mutations in the STIL gene are connected to microcephaly a condition characterized by underdeveloped cerebral cortex. Here STIL mutation affects proteins like MCPH1 which is involved in brain development.

Plays an important role in embryonic development as well as in cellular growth and proliferation; its long-term silencing affects cell survival and cell cycle distribution as well as decreases CDK1 activity correlated with reduced phosphorylation of CDK1. Plays a role as a positive regulator of the sonic hedgehog pathway, acting downstream of PTCH1 (PubMed : 16024801, PubMed : 9372240). Plays an important role in the regulation of centriole duplication. Required for the onset of procentriole formation and proper mitotic progression. During procentriole formation, is essential for the correct loading of SASS6 and CENPJ to the base of the procentriole to initiate procentriole assembly (PubMed : 22020124). In complex with STIL acts as a modulator of PLK4-driven cytoskeletal rearrangements and directional cell motility (PubMed : 29712910, PubMed : 32107292).
