Syntaxin 11 (STX11) is a member of the syntaxin family known as an important component in vesicle trafficking. This protein has a molecular mass of about 33 kDa. STX11 is mainly expressed in immune system cells including natural killer (NK) cells and macrophages. It plays a significant role in vesicle fusion processes by mediating the assembly and disassembly of SNARE complexes which are critical for vesicular transport and membrane fusion.
Biological function summary

Syntaxin 11 acts within the immune system to mediate secretory functions. It forms part of the SNARE complex and partners with other SNARE proteins to facilitate the exocytosis of cytotoxic granules from NK cells and cytotoxic T lymphocytes. This process is essential for immune cell-mediated destruction of infected or malignant cells. STX11 enables the regulation of immune responses by controlling vesicle docking and fusion during immune cell activation.

Pathways

The role of STX11 connects to critical immune response pathways. It plays a role in the regulation of the cytotoxic granule exocytosis pathway. This protein interacts with MUNC18-2 and other SNARE-related proteins facilitating vesicle transport and membrane fusion processes. STX11 is also involved in the modulation of pathways that oversee immune cell cytotoxicity and inflammatory responses.

Syntaxin 11 shows a connection to hemophagocytic lymphohistiocytosis (HLH) a severe immune dysregulation disorder. Alterations in STX11 have been linked to familial HLH where defective granule exocytosis impairs the immune system's ability to control inflammation leading to excessive immune activation. Interactions between STX11 and other proteins such as perforin are important in understanding HLH pathogenesis. Studies also suggest a potential connection to other immune system disorders though research continues to explore these relationships.
