4 ug/mL Succinic cysteine-crosslink-BSA was coated onto 96-wells. Serial dilution of free Succinic cysteine (0, 195.3125, 390.625, 781.25, 1562.5, 3125, 6250, 12500, 25000, 50000 ng/mL) was added (50 uL). This was followed by adding 0.1 ug/mL ab322069 (50 uL) into each well. The plate was incubated for 30 mins. After washing, 1 : 20000 diluted GAR Fc-HRP was added and incubated for 30 mins. After washing, 100 uL of TMB was added and incubated for 5-10 mins without shaking for color development. Then 100 uL of stop solution (4.9% Orthophosphoric acid) was dispensed and OD was read at 450nm on a microplate reader within 5-10 mins.

Succinic cysteine also known as S-cysteinylsuccinic acid is a modified amino acid and a derivative of succinic acid. It forms through the addition of a succinyl group to cysteine. The molecular mass of succinic cysteine is approximately 193.22 g/mol. It primarily accumulates in the mitochondria due to its involvement in metabolic processes. Succinic cysteine does not have expression in tissues or cellular compartments like conventional proteins but is more of a biochemical entity.
